The name of the priority area of science development for which the application is submitted 6. Life and health science.

The name of the specialized scientific research area: 6.1 Study, conservation and rational use of genetic resources of Kazakhstan. Innovative approaches to monitoring and protection of the environment. Impact of environmental factors on human health.

Scientific director of the project/program: Ulyanov Vadim, PhD.

The implementation period, its duration in months. 02.2023 - 12.2025 year, duration 35 month.

The total amount of funding, and by year: 75 000 thousand tenge,

2023 year – 25 000 thousand tenge;

2024 year – 25 000 thousand tenge;

2025 year – 25 000 thousand tenge.

Objects of research, development or design: The object of the study is Saiga tatarica tatarica from three Kazakh populations - Ural, Betpakdal and Ustyurt.

The purpose of the project / program: To study and characterise the allelofond of Kazakh populations of Saiga tatarica tatarica based on genome wide coverage SNP-genotyping data in the aspect of conservation of this species.

Project / program objectives:

1 take samples of biological material for molecular genetic testing;

2 Isolate DNA from selected biomaterial, assess the qualitative and quantitative parameters of the isolated DNA material;

3 Establishment of a depositary bank for the storage of saiga biomaterial and DNA;

4 Conducting genome wide genome-wide coverage SNP genotyping on wild populations and captive saigas;

5 Processing of SNP genotyping results (quality control and filtering of SNP markers, selection of polymorphic SNPs);

6 Mitochondrial genome sequencing and evolutionary vector studies, identifying rare and significant mutations in coding genes;

7 Assessment of genetic diversity and degree of genetic differentiation of Kazakh populations of Saiga tatarica tatarica;

8 Development of recommendations aimed at preserving the gene pool of Kazakh populations of Saiga tatarica tatarica.

The results obtained and the novelty: During the reporting period, the depository Bank was replenished with samples of the found biomaterial of the Ustyurt saiga population and the obtained DNA material. The sequencing results were aligned with the reference genome of Saiga tatarica, which contains about 2.9 billion base pairs collected in 1772199 scaffolds without annotations. The number of readings obtained for each sample ranged from 99,554,798 to 214,952,300. Of these data, 96.45% to 98.43% of the readings were successfully aligned to the reference genome. The most common substitution is A>G, and transitions prevail over transversions (ratio ≈ 2.18). The total length of the mitochondrial genome is ~16,300 base pairs, which indicates the preservation of the size of the mitochondrial genome. The distribution of coding sequences (CDS) is the same and occupies most of the outer ring in both genomes. Regions with high and low GC content show similar patterns, which indicates the conservativeness of the overall genome structure. The analyzed samples demonstrate generally high genetic similarity relative to each other based on mitochondrial genomes. However, the presence of genetic distance in the studied samples may indicate a heterogeneous population structure or geographical variation.

Expected results: The results of the work done will be as follows:

1 Genotyping will be carried out using genome wide coverage SNPs;

2 The results of SNP genotyping are processed;

3 The mitochondrial genome will be sequenced, the evolutionary vector will be studied, and rare and significant mutations in the coding genes will be identified;

4 The genetic diversity and degree of genetic differentiation of Kazakh populations of Saiga tatarica tatarica will be assessed;

5 Recommendations will be developed to preserve the gene pool of the Kazakh populations of Saiga tatarica tatarica.

Objects of research, development or design: The research will be carried out on a collection herd of waterfowl from a poultry farm in the Northern region of Kazakhstan.

The purpose of the program: Development of effective ways of breeding waterfowl through selection and technological methods and information technologies.

Program objectives:

To achieve this goal, the following tasks must be solved:

1. Create a new breed of meat ducks.

2. To develop feeding rations for ducks of different age and gender groups.

3. To develop information technologies for automation of the production process in the cultivation of waterfowl.

Today, the task is urgent not only to preserve the numerous diversity of lines, breeds, populations, but also their rational use, both for the production of eggs and meat, and for the creation of new more highly productive breeds and crosses.

Since the development of poultry farming will continue to be based on the use of hybrid poultry obtained as a result of crossing according to certain schemes, it is necessary to constantly work on increasing the heterosis effect based on the compatibility of breeds, crosses and lines.

The share of genotypic diversity in the total variability in most economically useful traits (egg production, precocity, live weight, etc.) is more fully manifested with proper cultivation and full-fledged feeding of poultry.

When growing and keeping linear and hybrid birds under the same conditions, it is possible to determine the relative value of a particular parent breed, line, cross in various crossing combinations and identify effective ways to use data to obtain a highly heterotic bird.

The efficiency of duck meat production largely depends on the payment of feed products, which is also considered an important task. It was believed that live weight selection was the simplest and most effective way to reduce feed costs, although the content of lean meat in the carcass may not increase.

Therefore, the choice of environmental conditions during the rearing of young animals plays an important role in improving the effectiveness of the assessment and selection of poultry for the reproduction of the herd. The "genotype—environment" interaction is defined in a two-factor dispersion complex as the proportion of the influence of a combination of genotypic and environmental factors.

In the structure of the cost of eggs and poultry meat, feed accounts for 60-70%. Reducing feed consumption per unit of production and increasing the efficiency of their assimilation can be achieved: firstly, by using high-quality feed products and proper preparation of feed for feeding; secondly, by using optimal feed recipes for the appropriate groups of poultry and, thirdly, by selecting appropriate feeding regimes and techniques.

Conducting economic analysis is an integral part of the work to optimize production activities and increase profits by improving production performance or by reducing costs.

Analyzing the production performance of a poultry farm is a difficult task. For its successful implementation, it is necessary to solve both the tasks of accounting for poultry stock and daily production indicators (poultry mortality, egg production, poultry slaughter, feed consumption and composition, transfer of poultry from the buildings of the repair young to the buildings of the parent herd, and others), and analysis of vaccination and prevention methods used at the poultry farm.

The purpose of automation, first of all, is to reduce the time spent by the building attendants and the dispatcher on calculating production indicators.

The technical level in the field of poultry farming in the Republic of Kazakhstan requires improvement.

The objectives of this project directly correspond to the strategic objectives of the Republic of Kazakhstan in the field of agriculture and food security. Improving the quality and efficiency of poultry production contributes to the development of the egg and meat industries and supports the country's strategic plans to provide food to the national market.

The results will meet the social demand for high-quality eggs and diurnal young, ensuring food safety and supporting the food industry. The economic and industrial interest in the results of the program is manifested in increasing the competitiveness of local producers and their ability to compete in the global market.

The results obtained and the novelty:

To increase the efficiency of poultry production, it is proposed to develop a digital small-sized machine in order to improve the quality and automatic sorting of incubation and commercial eggs. In addition, it is proposed to develop a sound stimulation device, the operation of which during the incubation of waterfowl eggs would ensure accelerated and synchronous withdrawal of daily young and would not only improve product quality, but also reduce human dependence and the likelihood of errors in the production process.

The task is an innovative solution that combines digital technologies with egg production, which makes it unique in the context of existing analogues. The fundamental difference lies in the development of a small-sized machine that not only determines quality indicators, but also automatically sorts eggs, which significantly increases the efficiency and accuracy of production processes.

Existing sorting and quality control methods remain largely manual and human-dependent. The task is a modern solution that will contribute to improving the technical level in this industry.

One of the reserves for increasing efficiency in poultry farming is considered to be the improvement of technological modes of egg incubation. In this direction, an important factor is to ensure accelerated and synchronous withdrawal of daily young in the incubator due to various stimulants, including sound ones.

Thus, automating the egg sorting and quality control processes will reduce the likelihood of human error, as well as ensure that eggs are sorted with high speed and accuracy. The use of a sound stimulation device during incubation will speed up the withdrawal of young animals from eggs.

Objects of research, development or design: estuaries

The aim of the project / program is to develop technologies for improving the ecological and meliorative state and increasing the productivity of estuaries

Project/program objectives:

1. Assessment of the ecological-hydrogeological-meliorative state of estuaries based on studies of soil indicators, the composition of the dominant taxa of microbiomes, hydrogeological and hydrochemical characteristics of groundwater, the regime and frequency of flooding, the qualitative composition of herbage.

2. Study of the influence of techniques for maintaining the water-salt regime of the soil and increasing the productivity of natural herbage on the ecological-hydrogeological-meliorative state of estuaries.

3. Substantiation of the influence of the landscape-ecological approach to the reclamation of estuarine lands on the maintenance of an optimal ecological-reclamation situation and restoration of the productivity of the natural herbage of degraded areas of the estuary.

The results obtained and the novelty:

More than 70% of engineering estuaries are flooded with violation of technological regimes.

Due to long-term flooding in the conditions of natural relief, an uneven impact on the landscape of the area occurs, which leads to a change in the water-salt balance of the territory, degradation of valuable plant species, and a decrease in the productivity of the natural herbage of estuaries.

Such a model of the development of secondary salinization processes was not considered during the design period and criteria for the intensity of the impact of estuarine irrigation on the formed estuarine agricultural landscape were not developed for these soil reclamation conditions.

These studies will be aimed at developing technologies to improve the ecological and meliorative state and increase the productivity of estuaries.

Currently, the use of modern methods of high-performance sequencing with the combination of traditional methods of soil science will allow a more informative study of the distribution of soil microorganisms to justify solutions to improve the ecological and reclamation state of agricultural landscapes.

The substantiation of the landscape-ecological approach to the reclamation of estuarine lands on the ecological-hydrogeological-meliorative state of estuaries, based on the established norms of the natural object, standardized parameters of the territory, the interval of their natural fluctuations, is a new level in scientific research, has significant scientific and technical potential.

The study of the microbial community on estuarine landscapes in conditions of long-term flooding, the change of which is an important symptom of violations of the natural processes of maintaining soil fertility, will have a significant impact on the level of research and scientific and technical potential.

The use of modern methods of high-performance sequencing with the combination of traditional methods of soil science in assessing the ecological and reclamation state of estuarine landscapes is a new and promising area of research.

Expected results:

Boudout:

- the influence of the flooding regime and chemical reclamation of estuary soils on the formation of the water-salt regime of the soil, as well as on the productivity of natural herbage, has been studied;

- а technology has been developed to improve the ecological and reclamation condition of estuaries and increase their productivity;

Also, formulations of feed mixtures will be developed and balanced feeding rations will be compiled.

The practical significance of the project lies in the fact that the conducted research will allow us to develop technological methods for intensifying meat production for high efficiency of meat production of poultry meat products, and significantly increase the profitability and competitiveness of the industry in Kazakhstan.

Research results:

The conditions of feeding and keeping of waterfowl in the West Kazakhstan region have been studied.

To complete the tasks, control (I, n =50/50) and experimental (II, n=50/50; III, n=50/50) groups of 100 heads each were formed. The birds were selected by the method of a group of analogues in terms of live weight and overall development. The live weight of daily ducklings ranged from 47.6 to 52.4 g. Differences in the live weight of ducklings of all groups at the age of 7 days were insignificant, and averaged 216.7 g.

Samples of the main types of concentrated feeds (soy, barley, wheat, rye, peas, wheat bran) were selected, and the nutritional value of grain feeds was analyzed. The research results showed that in grain feeds, namely soy, barley, wheat, rye, peas, the content of metabolic energy ranged from 9.8 to 13.2 MJ, and respectively amounted to 13.2; 11.0; 10.2; 9.8 and 10.5 MJ.

According to the results of the chemical composition of the feed, balanced formulations of feed mixtures for feeding experimental groups of ducks were compiled. Feeding of experimental groups of birds in all groups was carried out in two–phase periods: 1-14 and 15-49 days.

The content of crude protein in the diet of young animals of the III experimental group was comparatively higher than the peers of the I control and II experimental groups by 7.3 and 15.2%, respectively. In subsequent periods of growth (from the age of 15 days), Protein-vitamin-mineral concentrate (PVMC), a protein-feed mixture in different dosages, and a mineral composition additive were added to the diet.

As a result of production experiments, the data obtained on the dynamics of live weight in the studied groups of young ducks indicate that the addition of various feed additives to the diet has a positive effect on the productive indicators of birds.

Expected results:

Experimental groups of goslings of the Linda breed and ducks of the Medeo breed will be formed;

Balanced diets with the addition of biologically active additives will be compiled;

The chemical composition of the feed will be studied;

Formulations of feed mixtures for waterfowl of various sex and age groups will be developed;

The most optimal methods of intensification of geese cultivation technology will be determined;

The growth and development, safety, and productive indicators of experimental birds will be studied;

The digestibility of dietary nutrients in the body of experimental goslings and ducklings will be studied;

Hematological parameters of experimental goslings and ducklings will be studied;

The meat productivity of experimental goslings and ducklings will be studied;

An organoleptic assessment will be carried out and the morphological and chemical composition of the meat will be studied;

The economic efficiency of the intensification of the cultivation of geese and ducklings in the conditions of the West Kazakhstan region will be calculated.

The results obtained and the novelty: (no more than 200 words)

Zero tillage systems on seeders will allow you to add both seeds and seeds in combination with granular fertilizers. The method of applying granular fertilizers together with seeds is determined by the design of the coulters used by the zero tillage system. The distribution of granular fertilizers depends on the location of the seeds. The introduction of granular fertilizers and seeds to different depths is carried out by disc coulters. The technological process of the proposed sowing method is carried out by simultaneously embedding tuks of granular fertilizers to a greater depth than the seeds of the sown crops, i.e. a multi-level method of embedding tuks and seeds into the soil. The developed new two-disc coulter is designed for sowing seeds in a horizontal strip with a width of 12 cm, and granular fertilizers are 2 cm more than the specified depth of seed embedding and at the same time ensures the location of tuks in the middle of the strip.

Expected results:

1. Development of a working body with reasonable parameters that ensures the delay of seeds in the wavy seedbed after their exit from the seed duct.

2. Reasonable parameters of the disc coulter ripper having the ability to go deep to the desired depth and at the same time minimizing traction resistance.

3. Development of a six-row seeder for zero farming with a parallel mechanism for the working body.

The results obtained and the novelty: (no more than 200 words) Experimental groups of sturgeon fish have been formed, kept in conditions of closed-loop water supply installations. Subcutaneous microchips in a T-VAS Datamars needle were installed in selected sturgeon specimens (n=276). The initial biological material was selected for microsatellite analysis by taking ectodermal tissue – a fragment of the pectoral fin of sturgeons by the method of lifetime selection of biological samples, without using the decapitation technique. To qualitatively and quantitatively evaluate the isolated DNA from biological material, its concentration was measured on an Agilent Cary 60 spectrophotometer. The average DNA concentration was 156.8 ng/µl. STR-typing of 276 individuals of sturgeon fish contained in the conditions of ultrasound was carried out on the AB 3500 genetic analyzer (AppliedBiosystem, USA). A pair of primers were used to amplify each STR locus, providing effective and highly specific PCR amplification. DNA fragments were separated by capillary electrophoresis in a polymer gel. Based on the results of STR-typing, a genetic profile was compiled for each of the studied sturgeons.

Objects of research, development or design:

pyroplasmides, ixodides, cattle

The purpose of the project / program:

To study the current epizootic situation of piroplasmidosis and ixodidosis of cattle and to develop an effective treatment and prevention regimen for piroplasmidosis and ixodidosis in various natural zones of the West Kazakhstan region.

Project / program objectives:

1. To study the distribution and species composition of ixodid ticks in the steppe, semi-desert and desert zones of West Kazakhstan region. Ticks found on cattle will be identified to species. The distribution of ticks in various natural zones of West Kazakhstan region has been determined, which is necessary for the development of preventive measures.

2. To determine seasonal dynamics of tick infestation in cattle in various natural zones of West Kazakhstan region. To determine infestation of ixodids by piroplasmids using the PCR method. Seasonal dynamics, the index of tick abundance in cattle and the infestation of ixodids by piroplasmids, necessary for the development of preventive measures, will be determined.

3. To determine the age dynamics of ixodidae infestation in cattle in various natural zones of WK region and their distribution on the body of animals, taking into account the age structure (larva - nymph - imago).

4. To study epizootic situation regarding piroplasmosis in cattle in various natural zones of WK region. The extent of invasion of cattle by piroplasmids and their type will be determined by studying blood smears and PCR, necessary for the development of preventive measures.

5. To determine the nature of the course and symptoms of piroplasmosis in cattle in various zones of WK region. The nature of the course and symptoms of the disease necessary for the development of preventive measures will be determined.

6. To determine seasonal and age-related dynamics of piroplasmosis in cattle in various zones of WK region using the PCR method. Seasonal and age dynamics of piroplasmosis in cattle will be determined, which are necessary for the development of preventive measures.

7. To determine medication effectiveness against piroplasmosis. The effectiveness of anti-pyroplasmidose medication will be determined using PCR, a complex for pathogenetic therapy of cattle will be compiled, necessary for the development of preventive measures..

8. To study the effectiveness of acaricides used against ixodids. The effectiveness of acaricides will be determined by invitro and invivo methods, necessary for the development of preventive measures.

The results obtained and the novelty: (no more than 200 words)

The seasonal dynamics of infection of cattle with ticks was determined by quarterly (winter, spring, summer and autumn) animal studies, and age-based studies of various age groups: under one year, 1-3 years, 4-5 years, 6 years and older. The collection of ticks from vegetation was carried out on the "flag". The collection of ticks from cattle was carried out with tweezers in cans with a capacity of 100 mm3. In the steppe, semi-desert and desert zones of the West Kazakhstan region, cattle are infected with ticks of the Ixodidae family, belonging to five genera and ten species of ticks. The highest infection rate was observed in spring and summer and ranged from 100 to 90%. The greatest animal infestation was recorded in the steppe zone, and the lowest in the desert zone. Index of occurrence is averaged 55%, the highest index of abundance was determined in H. detritum – 59.3±12.1, and the lowest in I. ricinus - 3.5±1.0 ex/head. Young animals under the age of one year and from 1 to 3 years old were most affected, and the index of abundance was 216.2 - 252.4 ex/head, respectively.

Novelty: the development of a rational scheme for the treatment and prevention of pyroplasmidosis, and ways to control ticks.

Expected results:

1. Distribution and species composition of ixodid ticks in cattle in the steppe, semi-desert and desert zones of West Kazakhstan region will be determined.

2. Seasonal dynamics, tick abundance index in cattle and ixodid piroplasmid infestation will be determined using the PCR method.

3. Age dynamics of ixodid infestation in cattle, the distribution of ixodids on the body of animals, taking into account age structure (larva - nymph - imago) and the total stock of ixodids in various natural zones of West Kazakhstan region will be determined.

4. The extent of invasion of cattle by piroplasmids and their type will be determined by studying blood smears and PCR in various natural zones of West Kazakhstan region.

5. The nature of the course and symptoms of piroplasmosis in cattle in various zones of West Kazakhstan region will be determined.

6. Seasonal and age-related dynamics of piroplasmosis in cattle will be determined in various zones of West Kazakhstan region using the PCR method.

7. Comparative assessment of the effectiveness of the medication used against bovine piroplasmosis using PCR in the steppe, semi-desert and desert zones of West Kazakhstan region will be given.

8. Effectiveness of acaricides used against ixodid ticks using invitro and invivo methods will be determined.

9. Rational scheme will be developed and recommendations will be prepared for the prevention of piroplasmosis and ixodidosis in cattle in West Kazakhstan region.
